Unable to paint on transparent layer

I have a problem trying to paint on a transparent layer with a third party brush.

I have a project that has three visible layers. A white one at the bottom, a transparent one on the top, and picture on the intermediate layer. There are other layers, but they are not visible.

If I activate the bottom layer, I can paint on it, but if I activate the top transparent layer and try to paint, nothing happens that is visible. Layers are normal mode, opacity is 100%. Nothing is selected.

If I create a completely new, transparent, image I am able to paint on that.

I'd be grateful for advice on how to fix this. Is there some setting that I might have screwed up? Basically I'm wanting to paint over certain parts of the intermediate image, but keep the result as a separate layer.

Check the "Lock Alpha Channel". If it's pressed, the nothing will show on the layer.
